Most people do not keep the same job (or even career) for their entire working life. As time goes on in your job, certain things can wear you down. Your salary isn't quite making ends meet, your benefits don't match your needs and you're working the hours of a CEO despite your job title. Your job has gone from amazing to awful in a short period of time or your company has gone through a restructing. Either way, and once again, you're ready to join the rat race to look for a new one.
So as you can see, there are lots of reasons why people change jobs. However, how does one go about preparing for the big hop?

Elevator Speech - your speech should be around 30 seconds, focus on what you are looking for and what you have to offer. That you worked for xyz for 20 years doesn't really matter in a quick pitch. Consider your Elevator Speech as a "Mission Statement." You should be able to identify exactly what you are looking for; if you can not, you need to consider help in focusing where you want to be in 5 years.
